New large-scale research shows that genetics contribute meaningfully to personality alongside upbringing and life experience. Using the largest combined dataset to date, scientists identified 1,260 DNA variants associated with the Big Five personality dimensions: extraversion, agreeableness, conscientiousness, neuroticism and openness to experience.
Study Design and Scale
The meta-analysis pooled data from 46 independent cohorts and, depending on the trait analyzed, included between 611,037 and more than 1.13 million participants. Participants completed questionnaire-based personality measures and provided genome-wide genetic data, enabling researchers to test millions of small DNA differences against trait scores.

The study also connects genetic profiles for specific traits with life outcomes. For example, polygenic patterns associated with higher conscientiousness correlated with lower substance use, more participation in sports, fewer hospital visits, lower body mass index and markers of healthier aging. Genetic patterns tied to higher neuroticism were linked to anxiety- and depression-related conditions.
"There is no single 'personality gene,'" said Ted Schwaba, lead author and faculty member in the Department of Psychology at Michigan State University. "Personality traits are associated with many genetic variants, but even the largest effects of individual genetic variants are still very small."
Limitations and What It Means
The findings show that many common variants each exert tiny effects; when aggregated, their influence becomes meaningful but still explains only a modest fraction of individual differences. The heavy European-ancestry composition of the sample means further work is needed in more diverse populations to confirm and extend these results. The researchers and commentators emphasize that environmental factors and life experience remain powerful forces shaping personality.
The study, published in Nature, advances our understanding of how genes and environment interact to shape behavior and health. Better knowledge of these genetic associations may eventually inform personalized approaches to mental-health research and interventions, but it does not reduce personality to DNA alone.
